Methodology
Short tandem repeat (STR) polymorphic DNA markers are amplified by polymerase chain reaction (PCR) using the Promega PowerPlex 16 HS System, labeled with fluorescent markers and used to distinguish patient and donor cells as different on a Capillary Electrophoresis Fluorescence Detection instrument.
STR markers used in analysis include: D3S1358, TH01, D21S11, D18S51, Penta E, D5S818, D13S317, D7S820, D16S539, CSF1PO, Penta D, Amelogenin, vWA, D8S1179, TPOX, and FGA. (PCR is utilized pursuant to a license agreement with Roche Molecular Systems, Inc.)

Specimen Requirements
Call Barnes-Jewish Hospital Molecular Diagnostics Laboratory at 314-454-8685 for collection instructions.
Form: Molecular Diagnostics Requisition
Submit only 1 of the following specimens:
Specimen Type: Peripheral blood
Container/Tube: Lavender top (EDTA)
Specimen Volume: 2 mL
Specimen Minimum Volume: 1 mL
Collection Instructions: Invert several times to mix. Clotted blood is not acceptable.
Additional Information: Specimen cannot be frozen.
Specimen Type: Bone marrow
Container/Tube: Lavender top (EDTA)
Specimen Volume: 2 mL
Specimen Minimum Volume: 1 mL
Collection Instructions: Invert several times to mix bone marrow. Clotted bone marrow is not acceptable.
Additional Information: Specimen cannot be frozen.
Specimen Type: Cord blood, anticoagulated
Container/Tube: Lavender-top (EDTA)
Specimen Volume: 2 mL
Specimen Minimum Volume: 1 mL
Collection Instructions: Invert several times to mix cord blood. Clotted cord blood is not acceptable.
Additional Information: Specimen cannot be frozen.
Specimen Type: Buccal swabs
Call Barnes-Jewish Hospital Molecular Diagnostics Laboratory at 314-454-8685 for details about collection system and shipment. DNA, sufficient for PCR-based assays, may be isolated from cells obtained by a buccal swab; this is particularly well tolerated by children.
Specimen Transport Temperature
All specimen typesshould be sent at ambient or refrigerated temperatures.
